A sensitive, highly specific gas-liquid Chromatographic method for the quantitative determination of morphine in biological specimens is described. The quantitative method is based on the conversion of morphine to its diacetyl derivative, an approach which is preferable over the procedures that determine morphine as the free base. Preparation of the diacetyl derivative of morphine has several advantages of which a major one is that absolute dryness is not a requisite for quantitative formation of the required derivative. A rapid thin-layer Chromatographie technique for qualitative determination of morphine is also presented.
